Clearance by Macrophages of Alzheimer's Disease Patient - J Alzheimers Dis.

"1,25D3 strongly
stimulated Abeta phagocytosis and clearance while protecting against apoptosis.
Certain synthetic curcuminoids in combination with 1,25D3 had additive effects
on phagocytosis in Type I but not Type II macrophages. In addition, we
investigated the mechanisms of 1,25D3 and
curcuminoids in macrophages. The 1,25D3 genomic antagonist analog MK
inhibited 1,25D3 but not curcuminoid effects, suggesting that 1,25D3 acts
through the genomic pathway. In silico, 1,25D3 showed preferential binding to
the genomic pocket of the vitamin D receptor, whereas bisdemethoxycurcumin
showed preference for the non-genomic pocket. 1,25D3 is a promising hormone for
AD immunoprophylaxis because in Type I macrophages combined treatment with
1,25D3 and curcuminoids has additive effects, and in Type II macrophages 1,25D3
treatment is effective alone. Human macrophages are a new paradigm for testing

Curcumin
delays development of medroxyprogesterone acetate-accelerated
7,12-dimethylbenz[a]anthracene-induced mammary tumors - Menopause. 2009 Jul
22 - "Combined hormone
therapy (HT) containing estrogen and progestin (medroxyprogesterone acetate
[MPA]) leads to increased risk of breast cancer
in postmenopausal women, compared with HT
regimens containing estrogen alone or placebo ... Treatment with
curcumin delayed the first appearance of
MPA-accelerated tumors by 7 days, decreased tumor incidence by the end of the
experiment, and reduced tumor multiplicity in DMBA-induced MPA-accelerated
tumors. Curcumin also prevented many of the gross histological changes seen in
the MPA-treated mammary gland. Immunohistochemical analyses of mammary tumors
showed that curcumin decreased MPA-induced VEGF induction in hyperplastic
lesions, although it did not affect the levels of estrogen and progesterone

Curcumin
sensitizes human colorectal cancer to capecitabine by modulation of cyclin D1,
COX-2, MMP-9, VEGF and CXCR4 expression in an orthotopic mouse model - Int J
Cancer. 2009 May 26 - "Because of the poor prognosis and
the development of resistance against chemotherapeutic drugs, the current
treatment for advanced metastatic colorectal cancer
(CRC) is ineffective. Whether curcumin (a
component of turmeric) can potentiate the effect of capecitabine against growth
and metastasis of CRC was investigated ... In nude mice, the combination of
curcumin and capecitabine was found to be more effective than either agent alone
in reducing tumor volume (p = 0.001 vs. control; p = 0.031 vs. capecitabine
alone), Ki-67 proliferation index (p = 0.001 vs. control) and microvessel
density marker CD31. The combination treatment was also highly effective in
suppressing ascites and distant metastasis to the liver, intestines, lungs,
rectum and spleen. This effect was accompanied by suppressed expression of
activated NF-kappaB and NF-kappaB-regulated gene products (cyclin D1,c-myc,
bcl-2, bcl-xL, cIAP-1, COX-2, ICAM-1, MMP-9, CXCR4 and VEGF). Overall, our
results suggest that curcumin sensitizes CRC to the antitumor and antimetastatic
effects of capecitabine by suppressing NF-kappaB cell signaling pathway"

Omega-3
fatty acid supplements improve the cardiovascular risk profile of subjects with
metabolic syndrome, including markers of inflammation and auto-immunity -
Acta Cardiol. 2009 Jun;64(3):321-7 - "were given 1 gram
of fish oil as a single capsule, containing 180 mg
eicosapentaenoic acid and 120 mg
docosahexaenoic acid daily for 6 months. Control
subjects did not receive any supplementation over the same period. RESULTS: The
study was completed by 47 subjects in the intervention group and 42 subjects in
the control group. Treatment with omega 3 supplements was associated with a
significant fall in body weight (P < 0.05), systolic
blood pressures (P < 0.05), serum low density
lipoprotein cholesterol (P < 0.05), and total cholesterol (P < 0.05),
triglycerides (P < 0.05), high-sensitivity
C-reactive protein (hs-CRP) (P <
0.01), and Hsp27 antibody titres (P < 0.05). No significant changes were
observed in the control group. CONCLUSION: It appears that omega 3 improves the
cardiovascular risk profile of subjects with

progressively recovered toward baseline over 8 weeks posttreatment. Total and
high molecular weight adiponectin levels followed specular trends. Diastolic
blood pressure significantly decreased only in those with higher GDRs. Treatment

Letrozole once a week normalizes serum testosterone in obesity-related male

"Isolated hypogonadotropic hypogonadism (IHH) is
frequently observed in severely obese men, probably as a result of increased
estradiol (E(2)) production and E(2)-mediated negative feedback on pituitary
LH secretion. Aromatase inhibitors can reverse this process ... treated with
2.5 mg letrozole once a week for 6 months ... Six weeks of treatment reduced
total E(2) from 123+/-11 to 58+/-7 pmol/l (P<0.001, mean+/-s.e.m.), and
increased serum LH from 4.4+/-0.6 to 11.1+/-1.5 U/l (P<0.001). Total
testosterone rose from 5.9+/-0.5 to 19.6+/-1.4 nmol/l (P<0.001), and free
testosterone from 163+/-13 to 604+/-50 pmol/l (P<0.001). Total testosterone
rose to within the normal range in all subjects, whereas free testosterone
rose to supraphysiological levels in 7 out of 12 men ... Letrozole 2.5 mg

I've been taking a quarter table a day but it sounds like that's way to much
an a quarter tablet every other day might be best which comes to 2.5 mg
every 8 days. People complain about the hassle to get doctors to prescribe
testosterone. It sounds like letrozole will give you the same or better
numbers without the hassle. Plus testosterone replacement increases
estrogen but letrozole has the advantage of decreasing it. See my

Letrozole normalizes serum testosterone in severely obese men with
hypogonadotropic hypogonadism - Diabetes Obes Metab. 2005 May;7(3):211-5
- "Six weeks of treatment decreased serum estradiol
from 120 +/- 20 to 70 +/- 9 pmol/l (p = 0.006). None of the subjects
developed an estradiol level of less than 40 pmol/l. LH increased from 4.5
+/- 0.8 to 14.8 +/- 2.3 U/l (p < 0.001). Total testosterone rose from 7.5
+/- 1.0 to 23.8 +/- 3.0 nmol/l (p < 0.001) without a concomitant change in
sex hormone-binding globulin level. Those treated with Letrozole 17.5 mg per

aromatase inhibition: effects on glucose metabolism and serum leptin levels in
young and elderly men - Eur J Endocrinol. 2008 Dec 2 -
"Ten elderly and nine young healthy men were randomized
to receive letrozole 2.5 mg daily or placebo for 28 days in a crossover design.
Results: Both in young and elderly men, active treatment significantly increased
serum testosterone (+128 and +99%, resp.) and decreased estradiol levels (-41
and -62%, resp.). Fasting glucose and insulin levels decreased in young men
after active intervention (-7 and -37%, resp.) compared to placebo. Leptin
levels fell markedly in both age groups (-24 and -25%, resp.), while adiponectin
levels were not affected by the intervention. Lipid profile was slightly
impaired in both groups, with increasing LDL-cholesterol levels (+14%) in the
younger age group and 10% lower levels of ApoA1 in the elderly. A decline in
IGF-1 levels (-15%) was observed in the younger age group. No changes in weight

- It's something to think about for men taking anastrozole for
aromatization.

have read that Indole-3-Carbinol (I3C) actually increases the activity of
aromatase enzyme and that DIM does not - Life Extension Magazine, 3/01 -
"In one study, men received
I3C for one week; a profile of 13 estrogens was measured. The results
were that I3C significantly increased the urinary excretion of C-2
estrogens. The urinary concentrations of nearly all other estrogen
metabolites, including levels of estradiol, estrone, estriol and 16
alpha-hydroxyestrone, were lower after I3C treatment [J Natl Cancer Inst
1997 May 21;89(10):718-23]. On the other hand, studies have shown that
DIM can actually induce
aromatase activity with a two-fold increase [Toxicol Sci 2001
May;61(1):40-8]" - The following are the two references for that

- So if C-2 estrogens are increased, is that bad in men? I don't know.

metabolism proceeds by hydroxylation at one of two mutually exclusive
sites at C-2 and C-16. The catechol pathway yields the weakly estrogenic
2-hydroxyestrone (2-OHE1), which inhibits breast cell
proliferation" - If that means, and I think it does, that
the C-2 metabolites inhibit breast cell growth, that would definitely be
a good thing for men.
