Recent Longevity News for the week ending 6/9/21

'Remarkable' Response to Diabetes Drug in Resistant Bipolar Depression - Medscape, 6/7/21 - "The study needs replication, but this early clinical trial suggests that the mitigation of insulin resistance by metformin significantly improves depressive symptoms in a significant percentage of treatment resistant bipolar patients ... Patients were titrated up to 2000 mg of metformin, which was the full dose, over 2 weeks and then maintained on treatment for a further 24 weeks ... Those who reversed their insulin resistance showed a remarkable resolution in their depressive symptoms. The reduction in MADRS scores began at week 6, and were maintained through to the end of the study, and the Cohen's d effect size for MADRS depression scores for converters was 0.52 at week 14 and 0.55 at week 26 ... All had failed, on average, 8 or 9 trials in their lifetime. When they came to us, nothing else would work. That's one of the remarkable things about our results, just how well they responded when they had not responded to any other psychotropic medications" - See metformin at ReliableRX.

5 Ways to Keep Your Brain Sharp As You Age - Time, 6/7/21 - "As the body gets older, one of the best ways to keep it young is to stay physically active. The same is true for the brain. “If there’s only one thing you can do for brain health,” says Lock, “the evidence for exercise is overwhelming.” Working out increases a protein called brain-derived neurotrophic factor (BDNF), which is vital for growing and keeping neurons. Exercise also helps prevent brain inflammation, among other benefits. Strive for 150 minutes of aerobic workouts and one to two days of strength training each week ... After exercising, choose a brain-healthy recovery meal. In his research, Dr. Nikolaos Scarmeas, associate professor of neurology at Columbia University, found that the more closely adults followed a Mediterranean diet—having fish and plant-based foods such as fruit, vegetables, nuts and olive oil, while limiting red meat—the more their risk for Alzheimer’s disease dropped. The strictest followers cut their risk by 40%."

Anticholinergic Burden a Key Contributor to Cognitive Impairment in Schizophrenia - Medscape, 6/3/21 - "Anticholinergic medication burden from antipsychotics, antidepressants, and other psychotropics has a cumulative effect of worsening cognitive function in patients with schizophrenia"

Thyroid-Stimulating Hormone Levels and Incident Depression - Medscape, 6/3/21 - "Overall, low TSH levels (1st quintile) were associated with incident depression (adjusted RR = 1.36, 95% CI 1.02–1.81), remaining significant for women (adjusted RR = 1.64, 95% CI 1.15–2.33), but not for men. The same results were found when restricting analysis to euthyroid participants (adjusted RR = 1.46, 95% CI 1.08–1.99), also significant for women only (adjusted RR = 1.63, 95% CI 1.12–2.38)"

Many Sunscreens Have High Levels of a Carcinogen, Pharmacy Tells FDA - Medscape, 5/28/21 - "Valisure, an online pharmacy known for testing every batch of medication they sell, announced this week that they petitioned the FDA to recall 40 batches of sunscreens and after-sun products they say tested for high levels of the chemical benzene. The company tested 294 batches from 69 companies and found benzene in 27% ― many in major national brands like Neutrogena and Banana Boat. Some batches contained as much as three times the emergency FDA limit of 2 parts per million (ppm) ... Long-term exposure to benzene is known to cause cancer in humans"

Waking just one hour earlier cuts depression risk by double digits, study finds - Science Daily, 5/28/21 - "This suggests that if someone who normally goes to bed at 1 a.m. goes to bed at midnight instead and sleeps the same duration, they could cut their risk by 23%; if they go to bed at 11 p.m., they could cut it by about 40%" - Note: Here's my sleep formula.  It might be overkill but it works: 1000 mg of l-tryptophan (see tryptophan), 750 mg of GABA (see GABA), 200 mg of l-theanine (see l-theanine), 1 Valerian Nighttime (see valerian), 300 mg of relora (see relora, lowers cortisol), 300 mg of phosphatidyl serine (see phosphatidyl serine, lowers cortisol), 100 mg  of French oak extract (See French oak extract), 1300 mg of ashwaganda (See ashwagandha), 1 Nature's Way Chamomile Extract (see chamomile), tart cherry extract (see cherry extract), 88 mg saffron (see saffron), 25 mg B-6 - increases melatonin absorption, cut in half) (see B-vitamins), Jarrow B12/Folate and 3 mg melatonin (see melatonin).  Also, see my insomnia page.

Daily Cup of Coffee Cuts Type 2 Diabetes Risk by About 5% - Medscape, 5/27/21 - "Overall, an increase of one coffee cup a day was associated with a 4%-6% reduced risk of type 2 diabetes (hazard ratios, 0.94 for the Rotterdam Study and 0.96 for the UK Biobank study). The effects appeared strongest in drinkers of filtered or ground coffee vs. those who reported drinking mainly instant coffee ... Also, an increase in coffee consumption of one cup a day was linked to lower levels of longitudinally assessed homeostatic model assessment of insulin resistance (HOMA-IR), with lower C reactive protein (CRP) and higher levels of adiponectin ... Although the associations between coffee and type 2 diabetes have been previously reported, "this study offers important findings due to the carefully standardized analyses on these two major data sources," ... But what makes this study different is that "these investigators hypothesized that this association could be due to an anti-inflammatory benefit" -

Three years younger in just eight weeks? A new study suggests yes! - Science Daily, 5/27/21 - "The study, released on April 12, utilized a randomized controlled clinical trial conducted among 43 healthy adult males between the ages of 50-72. The 8-week treatment program included diet, sleep, exercise and relaxation guidance, and supplemental probiotics and phytonutrients, resulting in a statistically significant reduction of biological age -- over three years younger, compared to controls" - Note: "sleep".  See my sleep formula above.

For men, low testosterone means high risk of severe COVID-19, study suggests - Science Daily, 5/25/21 - "The lower the levels of testosterone, the more severe the disease. For example, those with the lowest levels of testosterone in the blood were at highest risk of going on a ventilator, needing intensive care or dying ... In addition, the researchers found that lower testosterone levels in men also correlated with higher levels of inflammation"

Amazon indigenous group's lifestyle may hold a key to slowing down aging - Science Daily, 5/26/21 - "the Tsimane have little or no access to health care but are extremely physically active and consume a high-fiber diet that includes vegetables, fish and lean meat ... the difference in brain volumes between middle age and old age is 70% smaller in Tsimane than in Western populations. This suggests that the Tsimane's brains likely experience far less brain atrophy than Westerners as they age; atrophy is correlated with risk of cognitive impairment, functional decline and dementia"

Researchers discover fundamental roles of glucosamine in brain - Science Daily, 5/26/21 - "Using biochemical approaches, the researchers determined the sugar composition of glycogen in the muscle, liver, and brain of mice. Unlike muscle glycogen, which had only 1% glucosamine, and liver glycogen, which had less than 1% glucosamine, brain glycogen contained 25% glucosamine" - See glucosamine hydrochloride at Amazon.com.

Depression and Vegetarians: Association between Dietary Vitamin B6, B12 and Folate Intake and Global and Subcortical Brain Volumes - Nutrients 2021 May 24 - "Deficiency of vitamin B6 and vitamin B12, mostly in vegetarians, is found to be associated with depression and adverse neurological function. We investigated whether vitamin B6, B12, and folate have an effect on brain structure, especially among depressed people who follow a specific diet ... Depression was more prevalent, and intake of vitamin B6 and B12 was lower among vegetarians, while non-vegetarians had a lower intake of folate. Overall, no significant association was observed between vitamin B6, B12, and folate intakes and both global and subcortical brain volumes among participants with depression. However, vitamin B12 intake was positively associated with right pallidum among non-depressed participants, and a significant interaction between vitamin B12 intake and depression status on the right pallidum was observed. Also, a significant interaction between folate intake and depression status on grey matter (GM) volume and left thalamus was observed. Upon diet stratification, folate intake is associated with total brain volume and GM volume among vegetarians with depression. Furthermore, no significant associations were observed for subcortical regions. Our findings suggest that dietary intake of vitamin B6 and B12 might have an effect on brain structure. Vegetarians, particularly those who suffer from depression may benefit from supplementing their diets with vitamins B6, B12, and folate to ensure brain health" - See B complex supplements at Amazon.com.

Luteolin and its derivative apigenin suppress the inducible PD-L1 expression to improve anti-tumor immunity in KRAS-mutant lung cancer - Cancer Lett 2021 May 27 - "Upregulated expression of immune checkpoint molecules correlates with exhausted phenotype and impaired function of cytotoxic T cells to evade host immunity. By disrupting the interaction of PD-L1 and PD1, immune checkpoint inhibitors can restore immune system function against cancer cells. Growing evidence have demonstrated apigenin and luteolin, which are flavonoids abundant in common fruits and vegetables, can suppress growth and induce apoptosis of multiple types of cancer cells with their potent anti-inflammatory, antioxidant and anticancer properties ... Luteolin and apigenin significantly inhibited lung cancer cell growth, induced cell apoptosis, and down-regulated the IFN-γ-induced PD-L1 expression by suppressing the phosphorylation of STAT3. Both luteolin and apigenin showed potent anti-cancer activities in the H358 xenograft and Lewis lung carcinoma model in vivo, and the treatment with monoclonal PD1 antibody enhanced the infiltration of T cells into tumor tissues. Apigenin exhibited anti-tumor activity in Genetically engineered KRASLA2 mice. In conclusion, both apigenin and luteolin significantly suppressed lung cancer with KRAS mutant proliferation, and down-regulated the IFN-γ induced PD-L1 expression. Treatment with the combination of PD-1 blockade and apigenin/luteolin has a synergistic effect and might be a prospective therapeutic strategy for NSCLC with KRAS-mutant" - See luteolin at Amazon.com.

Therapeutic potential of mega-dose vitamin C to reverse organ dysfunction in sepsis and COVID-19 - Br J Pharmacol 2021 Jun 1 - "Sepsis induced by bacteria or viruses can result in multi-organ dysfunction, which is a major cause of death in intensive care units. Current treatments are only supportive and there are no treatments that reverse the pathophysiological effects of sepsis. Vitamin C has antioxidant, anti-inflammatory, anticoagulant and immune modulatory actions, so is a rational treatment for sepsis ... Mega-dose intravenous sodium ascorbate (150 g/40 kg over 7-h) dramatically improved the clinical state and cardiovascular, pulmonary, hepatic and renal function and decreased body temperature, in a clinically relevant ovine model of gram-negative bacteria-induced sepsis. In a critically ill COVID-19 patient, intravenous sodium ascorbate (60 g) restored arterial pressure, improved renal function and increased arterial blood oxygen levels. These findings suggest that megadose vitamin C should be trialled as a treatment for sepsis and COVID-19" - See American Health Ester-c With Citrus Bioflavonoids 500 Mg, 240 Count.

Curcumin's Effect on Inflammatory Response following Percutaneous Coronary Intervention in Adult Patients with Stable Coronary Heart Disease - Int J Angiol 2021 Jun - "Cardiovascular diseases play major roles in the health problems worldwide especially in Indonesia. Percutaneous coronary intervention (PCI) is a minimally invasive procedure with relatively low complications. However, high inflammatory response post-PCI has showed adverse events even after administration of standard medication. Previous studies showed that curcumin was able to reduce inflammatory response in adult patients with stable coronary heart disease (CHD) ... Either curcumin (45 mg/day) or placebo was given 7 days prior to PCI until 2 days after PCI ... Curcumin significantly reduce the serum hsCRP ( p = 0.006) and sCD40L ( p = 0.002) 7 days before PCI to 48 hours post-PCI. The decrement of hsCRP (-14.2% vs. -7.4%) and sCD40L (-24.3% vs. -13.2%) from 24 to 48 hours post-PCI was higher in the curcumin group than placebo group. The administration of curcumin 45 mg dose daily for 7 days prior PCI until 48 hours post-PCI is useful in reducing inflammatory response post-PCI with stable CHD" - See curcumin at Amazon.com.

Pleiotropic nature of curcumin in targeting multiple apoptotic-mediated factors and related strategies to treat gastric cancer: A review - Phytother Res 2021 May 24 - "Gastric cancer (GC) is one of the major reasons for cancer-associated death and exhibits the second-highest mortality rate worldwide. Several advanced approaches have been designed to treat GC; however, these strategies possess many innate complications. In view of this, the upcoming research relying on natural products could result in designing potential anticancer agents with fewer side effects. Curcumin, isolated from the rhizomes of Curcuma longa L. has several medicinal properties like antiinflammatory, antioxidant, antiapoptotic, antitumor, and antimetastatic. Such pleiotropic nature of curcumin impedes the invasion and proliferation of GC by targeting several oncogenic factors like p23, human epidermal factor receptor2 including Helicobacter pylori. The side effect of chemotherapy, that is, chemotherapeutic resistance and radiotherapy could be reduced combination therapy of curcumin. Moreover, the photodynamic therapy of curcumin destroys the cancer cells without affecting normal cells. However, further more potential studies are required to establish the potent efficacy of curcumin in the treatment of GC" - See curcumin at Amazon.com.

Effect of probiotic supplementation along with calorie restriction on metabolic endotoxemia, and inflammation markers in coronary artery disease patients: a double blind placebo controlled randomized clinical trial - Nutr J 2021 Jun 1 - "Alterations in the gut microbiome (dysbiosis) has been associated with increased microbial translocation, leading to chronic inflammation in coronary artery disease (CAD). It has been proposed that modulation of gut microbiota by probiotic might modify metabolic endotoxemia ... These data provide preliminary evidence that probiotic supplementation has beneficial effects on metabolic endotoxemia, and mega inflammation in participants with CAD" - See probiotic supplements at Amazon.com.

Theacrine, a Potent Antidepressant Purine Alkaloid from a Special Chinese Tea, Promotes Adult Hippocampal Neurogenesis in Stressed Mice - J Agric Food Chem 2021 Jun 1 - "Daily intake of tea has been known to relate to a low risk of depression. In this study, we report that a special variety of tea in China, Camellia assamica var. kucha (kucha), possesses antidepressant effects but with less adverse effects as compared to traditional tea Camellia sinensis. This action of kucha is related to its high amount of theacrine, a purine alkaloid structurally similar to caffeine ... Results of behavioral and neurotransmitter analysis showed that intragastric administration of theacrine significantly counteracted chronic stress-induced depression-like disorders and abnormal 5-hydroxytryptamine (5-HT) metabolism with less central excitability. Further investigation from both in vivo and in vitro experiments indicated that the antidepressant mechanism of theacrine was associated with promoting adult hippocampal neurogenesis, via the modulation of the phosphodiesterase-4 (PDE4)/cyclic adenosine monophosphate (cAMP)/cAMP response-element binding (CREB)/brain-derived neurotrophic factor (BDNF)/tropomyosin-related kinase B (TrkB) pathway. Collectively, our findings could promote the prevalence of kucha as a common beverage with uses for health care and contribute to the development of theacrine as a potential novel antidepressant medicine" - See theacrine at Amazon.com.

Effects of chromium supplementation on oxidative Stress biomarkers - Int J Vitam Nutr Res 2021 May 20 - "Overall, this meta-analysis demonstrated that chromium supplementation increased GSH without any significant changes in the mean of GPX, MDA, TAS, TBARS, CAT and SOD" - See chromium supplements at Amazon.com.

The findings for use of statins with aggressive PCa are consistent with cohort studies"

Effects of alpha-lipoic acid treatment on serum progranulin levels and inflammatory markers in diabetic neuropathy - J Int Med Res 2021 May - "Progranulin (PGRN) is a secreted growth factor that helps to regulate neuronal survival by blocking tumor necrosis factor-alpha (TNFα) receptors. The antioxidant alpha-lipoic acid (ALA) is used in diabetic neuropathy to improve nerve conduction and relieve neuropathic pain, but its effects on PGRN levels have not yet been elucidated ... 54 patients with type 2 diabetes and peripheral neuropathy received 600 mg of ALA daily for 6 months. Twenty-four patients with diabetes without neuropathy were also included in the study ... After ALA treatment, serum PGRN levels were significantly increased and CPT values were significantly improved. Furthermore, there were significant positive correlations among TNFα, ICAM-1, and PGRN levels both before and after ALA treatment. A significant negative correlation was observed between the improvements in CPT and the PGRN levels. Furthermore, ICAM-1 levels were an independent predictor of PGRN levels ... Conclusions: Changes in serum PGRN levels indicate that ALA treatment may have beneficial effects on endothelial function and neuronal inflammation" - See alpha lipoic acid at Amazon.com.

Habitual coffee and caffeinated beverages consumption is inversely associated with arterial stiffness and central and peripheral blood pressure - Int J Food Sci Nutr 2021 May 31 - "Aortic stiffness, wave reflections, and central and peripheral blood pressure (BP) are milestone indicators of cardiovascular-risk ... A linear inverse relationship between coffee and caffeine consumption and arterial stiffness and central and peripheral BP was found. Light coffee and caffeine consumers showed β-coefficients for PWV-0.15, SBP-3.61, DBP-2.48, cSBP-3.21, and cDBP-2.18 (all p values < 0.05). Present findings suggest that coffee and caffeine consumption is inversely associated with arterial stiffness and central and peripheral BP in a large population sample"

Functional Characterization of Undenatured Type II Collagen Supplements: Are They Interchangeable? - J Diet Suppl 2021 Jun 1;1-16 - "Undenatured (native) type II collagen is a dietary supplement ingredient reported to support joint health in healthy individuals by providing relief from symptoms of stiffness and discomfort and improving mobility. This benefit is thought to occur through oral tolerance, a mechanism whereby the immune system distinguishes between innocuous material in the gut and potentially harmful foreign invaders. The presence of antigenic epitopes in undenatured type II collagen, but not in denatured (hydrolyzed) collagen, is thought to be the basis for the therapeutic benefits ... Collagen type II supplement powders (raw material) and capsules (products in the market) were examined for color, particle size, quality profiles, fatty acid profiles, electron microscopy, and were analyzed for amino acid content as well as antigenic potential via an ELISA assay. Powders labeled as undenatured type II collagen were found to have markedly different properties, including the size of collagen fibers as per electron microscopy and antigenic configuration as per the ELISA assay. As significant differences were found between products, it allows consumers and practitioners to not assume that products labeled as undenatured (native) type II collagen are interchangeable." - See NOW Supplements, UC-II Type II Collagen with Undenatured Type II Collagen, 120 Veg Capsules.

l -Arginine Can Enhance the Beneficial Effect of Losartan in Patients with Chronic Aortic Regurgitation and Isolated Systolic Hypertension - Int J Angiol 2021 Jun;30(2) - "chronic aortic regurgitation (CAR) and isolated systolic hypertension (ISH) ... Both groups had a significant reduction in systolic blood pressure (SBP) and diastolic blood pressure (DBP), left ventricular end-diastolic volume index (LVEDVI), LV end-systolic volume index (LVESVI), LV mass index (LVMI), and LV mean wall stress after 6- and 12-month treatment ( p <0.01 in all comparisons). Both groups had a significant increase in LV ejection fraction and exercise duration after 6- and 12-month treatment ( p < 0.01 in all comparisons). Using multivariate linear regression analysis, only losartan + l -arginine therapy achieved a significantly lower LVESVI (38.89 ± 0.23 mL/m 2 ), LVEDVI (102.3 ± 0.3 mL/m 2 ), LVMI (107.6 ± 0.3 g/m 2 ), SBP (123.5 ± 1.0 mm Hg), and greater exercise duration (7.38 ± 0.02 minutes) than those of the losartan-only treated groups ( p <0.01 in all comparisons). These findings suggest that early co-administrative strategy provides a beneficial approach to favorably influence the natural history of CAR" - See l-arginine products at Amazon.com.
